Buy your tickets online!!! This is how you'll get your time slot and immediately in without waiting. Otherwise you'll wait in the line for 2-3 hours. Online tickets are sold two months in advance, so you need to plan your visit quite early (maybe you'll be lucky as I was and you'll find a couple of available tickets while in Amsterdam, but don't take that risk). About the museum: What an emotional and touching visit. A true museum with a story (among other crappy Amsterdam museums and attractions). I highly recommend you to know at least something about Anne Frank and her life before going there (read her diary or watch one of the films about her life).
