Lines are very long. I suggest visiting the Sistine Chapel first before going to the Museum. More tourists go to the museum and then Sistine Chapel, going the other way will minimize wait times. In addition, you can purchase your tickets online before you visit. You can also going earlier in the morning to avoid bigger crowds.
