Definitely get the "skip the line" passes and go early in the morning (8am). There is quite a bit of walking and some stairs before you see the Sistine Chapel. They do allow photographs everywhere but the Sistine Chapel. You can easily spend hours in the museum and the church. We were able to see the old apartments that the Popes used and original works from Michelangelo and Raphael. You do have to pass through a metal detector to enter the museums and they will make you lock up large bags and umbrellas.
