Fantastic museum, but gets very crowded. After waiting 1 1/2 hours to get into the Colosseum earlier in the week we decided to get skip the line tickets for 8 am. Definitely worth the investment as the general admission queue was already large well before opening time. You can also get directly to St Peter's if you go through the right hand door at the end of the Sistine Chapel. Officially this door is for groups only, but it didn't seem to be tightly enforced, so you could just follow a tour group through.
